The WTF moment for me while reading through the mRNA Vaccine literature was when they started discussing the self adjuvant properties of the LNP package for the mRNA vaccines. The way the nano particles trigger the immune system. Or rather the effect happened but they had no real idea of the actual mechanisms involved.
From reading the rest of the vaccine literature it became obvious that only very few adjuvants are approved for mass use vaccines precisely because only a few have a large body of field and clinical data going back over decades that indicate the way the interact with the immune system and long term effects. Even so the preferred vaccines from a product safety point are those that dont need adjuvants.
The overwhelming impression I got from the current research of how mRNA-LNP interacts with the immune system short term let alone long term is that it could be useful for very specific low use / high risk scenarios (like an ebola) but for mass vaccination for a low health risk endemic infectious disease it was utterly irresponsible to use when so little is known about the interactions with the immune system quite separate from the delivery of any vaccine package.
This is one very serious problem that the adenovirus and attenuated virus vaccines dont have.
